In this study, rosuvastatin was evaluated in mice with hyperlipidemia, and its effect was assessed in terms of lipid profile and the expression of HMG-CoA- reductase. Additionally, lipid-lowering capacity of rosuvastatin in mice with hypercholesterolemia was investigated, as it may influence the therapeutic response and overall clinical outcome.
This study was approved by the Research Ethics Committee of the Sichuan University (Approval No. 2018-0004), and was registered in the Chinese Medical Journal (CJCMJ0002021).
All the animals were obtained from animal care and management committee at the University of Shanghai. Male C57BL/6 mice (5-6 week-old) were purchased from Vital River Laboratory (Wuhan, China). The animals were fed standard chow diet (LD12, LD10, and LD20) at normal or high-fat diet. The animals were treated daily with rosuvastatin (1000mg/day) for 3 weeks. In addition, the animals were kept under laboratory conditions with a 12 h light-dark cycle, and the diet contained natural organic and natural components, including cornified flour, tap water, cotton seed, walnuts, flaxseeds, and flaxseeds. All the experimental procedures were carried out in accordance with the approved guidelines.
To detect lipid levels, the mice were anesthetized with intraperitoneal ketamine (100mg/kg), and they were then sacrificed by cervical dislocation. The liver samples were collected, and the liver was collected in the same manner as those used for analysis. The liver was lysed in lysate buffer (100mL) containing the lipid peroxidation inhibitor p-nitrophenyl-glycerol, and the cell lysate was subjected to further analysis. Then, the enzyme-linked immunosorbent assay (ELISA) (Abcam, Inc., USA) was used to detect lipid levels in liver. The mice were then anesthetized with 4% isoflurane, and the abdomen was opened. The abdomen was then closed and the heart was isolated and collected, and then the liver was removed, and the liver was weighed. Finally, the liver was separated from the abdomen, washed by centrifugation, and suspended in distilled water and then placed in a water bath for 30 min at room temperature. The supernatant was collected and used for lipid profile analysis. All the other chemicals were purchased from China Chemical Society (Shanghai, China).
To our knowledge, this is the first report that rosuvastatin can effectively reduce the lipid-lowering capacity of a statin in mice with hyperlipidemia. We found that rosuvastatin had a lower lipid-lowering capacity than Crestor. Furthermore, lipid-lowering capacity of rosuvastatin was decreased in mice with hypercholesterolemia, which was consistent with our previous study [
